Multiplex Ligation-Dependent Probe Amplification
The target sequence in genomic DNA is hybridized to two adjacent complementary probes. By a subsequent ligation reaction, these two probes are joined. A primer pair is designed matching to one probe each. Amplification by this primer pair only occurs if the probes are joined, which requires them to hybridise properly. This reaction can be performed with many probe and primer pairs simultaneously in a multiplex reaction.
